Louis de Groot was a son of Mozes de Groot and Sientje Nieweg. Hij had three other brothers and a sister: Samuel, Marcus, Estella and Jozef. Louis married on 7 August 1924 in Groningen to Hennij Cossen. The couple had two children, Maurits and Martha and they took care of an orphant boy Iwan de Vries, who lived with them.
