Marcus Rubens was a son of Jacob Meijer Rubens and Esther Zadoks. He married Rebecca Witstein in Rotterdam on 8 March 1911, a daughter of Ezechiel Witstein and Hester Benedictus. The couple had three daughters, who all have survived the Holocaust and a son Jakob Gijs, who was killed in 1941 in Mauthausen.

Marcus Rubens and his wife had a wholesale business in buttons, ties and pins. Shortly after Marcus and Rebecca received the news of the death of their son 'Jaap', their maidservant urged them to go into hiding.
